Casimero began his professional campaign in June 2007. He won his first world title in 2012, stopping Luis Alberto Lazarte in Argentina for the IBF Interim Junior Flyweight Title in a fight that ended in a ring riot. He was later elevated to full champion.

He became a two-division champion in 2016 in a rivalry with Amnat Ruenroeng, winning the IBF Flyweight title via a fourth-round knockout.

In November 2019, he became a three-division champion by scoring a major upset TKO victory over Zolani Tete to win the WBO Bantamweight Title. He defended this title in a high-profile, low-action rivalry against Guillermo Rigondeaux, winning a split decision.
